Screening of Medium Components for Polyribosyl Ribitol Phosphate Production by Haemophilus Influenzae Type-b Using Plackett-burman Design

ثبت نشده
چکیده

Plackett-Burman experimental design was used to screen important medium components influencing the production of polyribosyl ribitol phosphate by Haemophilus influenzae type-B under submerged fermentation. Eleven media parameters such as Glutamic acid, Yeast extract, cystein, dextrose, NAD, heamin, NaCl, NH4Cl, Na2HPO4, KCl, and MgSO4.7H2O were screened in twelve experimental runs as per the design. From the standard Plackett-Burman data analysis it was conformed that, heamin, yeast extract, NAD, Na2HPO4 and dextrose had contributed to a large extent, MgSO4 and L-cystein had little impact, while, glutamic acid, NH4Cl and KCl contributes moderately for polyribosyl ribitol phosphate production by haemophilus influenzae type-B under submerged fermentation.

A study on Haemophilus influenzae type b growth rate and capsule production in different media

In the present study, seven isolates of Haemophilus influenzae type b were cultured in four different media to compare growth rate and capsule production. Four liquid media namely brain heart infusion broth (BHI), trypticase soy broth (TSB), Mueller Hinton Broth (MHB) and Gonococci broth (GC) with added supplements (1% hemoglobin, 1% Isovitalex) were used. The growth was measured by colony coun...

Improvement of Large-scale PRP production by Haemophilus influenzae typeb, using modified CY medium

Background and Objective: Haemophilus influenzae type b (Hib) is a gram negative bacterium and one of the most common causative agents of acute meningitis in infants and less than 5 years old children worldwide. The production of Hib capsular polysaccharide polyribosyl ribitolphosphate (PRP) is important for the production of conjugate vaccines against Hib infections. The aim of this study is t...

A modular synthetic route to size-defined immunogenic Haemophilus influenzae b antigens is key to the identification of an octasaccharide lead vaccine candidate

The first glycoconjugate vaccine using isolated glycans was licensed to protect children from Haemophilus influenzae serotype b (Hib) infections. Subsequently, the first semisynthetic glycoconjugate vaccine using a mixture of antigens derived by polymerization targeted the same pathogen. Still, a detailed understanding concerning the correlation between oligosaccharide chain length and the immu...

Avidity of specific IgG antibodies elicited by immunisation against Haemophilus influenzae type b.

AIM To investigate the avidity of specific IgG polyribosyl ribitol phosphate (PRP) antibodies induced by three Haemophilus influenzae type b (Hib) conjugate vaccines: PRP-meningococcal outer membrane protein complex (PRP-OMP), PRP-non-toxic mutant diphtheria toxin, CRM197 (HbOC) and PRP-tetanus toxoid (PRP-T). METHODS One hundred and ten infants were immunised with one of the vaccines at two,...

Immunisation of infants against Haemophilus influenzae type b in the UK.

Haemophilus influenzae is one of the microorganisms that make up the commensal flora of the upper respiratory tract. Carriage is common, but compatible with good health. None the less, H influenzae is a pathogen and, in the case of encapsulated type b organisms, causes life threatening, invasive (bacteraemic) infections, the most common and serious being meningitis and epiglottitis (table 1).' ...
